ABSTRACT The procedures for a set of techniques to recover grains of pollen 26 and spores from liquid samples are described. Liquefied samples of matrices, such as melting lakes, snow and ice, can provide a record of climatic and environmental events at different geographic and temporal scales. The need for standardization of techniques was demonstrated in research carried out with matrices of snow and water of thawing lakes and snow collected on King George and Joinville islands (Antarctica). The use of the methods described can permit increased rescue of palynomorphs from liquid matrices, thereby adding greater reliability to palynological data and its use as a biotracer of environmental events. ABSTRACT In order to successfully establish themselves in their natural environment, ferns need habitats with abiotic conditions that are suitable for spore germination and gametophyte development. The objective of this study was to assess the influence of abiotic factors on the initial development of Cyathea phalerata cultivated in vitro. Spore germination and gametophyte development were assessed under varying conditions of surface sterilization, pH, temperature and photoperiod. To study adhesion and viability of uredospores of the fungus Phakopsora pachyrhizi on soybean seeds during storage, suspension tests of those uredospores were carried out by washing seeds at each 30 days interval. Furthermore, germination and inoculation tests of uredospores on soybean plants were performed with uredospores collected from seeds of two soybean production areas, located in the municipalities "Chapada dos Guimarães" and "Tangará da Serra", State of Mato Grosso, Brazil. High levels of uredospores infestation were detected before storage [249.31 and 85.18 uredospores/100 seeds (U/100)] on seeds collected in both localities, respectively. Los hongos micorrízicos arbusculares (HMA) forman una asociación simbiótica con las raíces de las plantas, favoreciendo un incremento en la asimilación de nutrientes del suelo. Los HMA colonizan la mayoría de los cultivos agrícolas; entre ellos, el cultivo del papayo presenta alto grado de dependencia micorrízica. This paper presents morphological descriptions and ecological data of cryptogam spores and other non-pollen palynomorphs from Quaternary sediments of Campos Basin, Rio de Janeiro, SE Brazil. The ages were derived from biostratigraphy of planktonic foraminifers and two radiocarbon dates, and suggest that sediment deposition started in the last 140,000 years BP. Thirty different types of palynomorphs were identified, described, and photographed: two bryophyte spores (sensu lato); 21 pteridophyte spores; four freshwater microalgae; onePseudoschizaea; and two microfungi. Comparative palynological analysis of Lygodium venustum Sw. and L. volubile Sw. (Lygodiaceae) Anais da ABC (AABC)

The genus Lygodium Sw. is one of the few climbing ferns in the world. The spores of L. venustum Sw. and L. volubile Sw. were studied using light, scanning, and transmission electron microscopy. This work is based on herbarium material. The spores are trilete, triangular, with straight to convex sides in polar view. The equatorial diameter is 72-104 µm, and the polar diameter is 64-84 µm. The ornamentation in L. venustum is verrucate-tuberculate while in L. volubile, it is verrucate-tuberculate in the proximal face but with a few ridges on the distal face, where a micro-ornamentation constituted by verrrucae and tubercles is observed. ABSTRACT: The objective of this study was to evaluate the effect of the initial microbial load, temperature and contact time on the biofilm formation of Alicyclobacillus acidoterrestris on stainless steel and natural food-grade rubber using orange juice as culture medium. The low initial load of A. acidoterrestris (2 log CFU/mL) led to biofilm formation on the stainless steel surface after 48 h of contact at 28 ºC and after 24 h at 45 ºC, and on natural food-grade rubber surface after 48 h of contact at both temperatures. Abstract: The genus Dermocystidium is very comprehensive in the host and site of infection, however this is the first report of the occurrence of Dermocystidium sp. in the gills of Nile tilapia. This study was carried out in a fish farming located in the state of Santa Catarina, Brazil. No mortalities were reported in the facility studied and the animals were clinically healthy. During the histopathological analysis of the gills, 8.33% of the fish presented spores of Dermocystidium sp. in the gill tissue. Avaliacao da producao de cupuacu (Theobroma grandiflorum) em diferentes sistemas agroflorestais (Sistema 1- seringueira x pupunha x mamao x cupuacu; Sistema 2- castanha-do-brasil x pupunha x urucum x cupuacu; Sistema 3- laranja x seringueira x coco x parica x cupuacu; e Sistema 4- monocultivo de cupuacu) em um experimento implantado na area experimental do CPAA, onde as plantas em parcelas cobertas por pueraria foram submetidas a dois niveis de adubacao (100% da adubacao recomendada e 30% da recomendada combinada com (C) e sem (S) inoculacao das plantas com FMVA). De acordo com os resultados parciais, os tratamentos nao exerceram influencia na producao de frutos. As plantas do Sistema 1 apresentaram maior producao que as dos demais sistemas.

Lab-Scale production of Bacillus atrophaeus' spores by solid state fermentation in fifferent types of bioreactors BABT

Studies were conducted to evaluate Bacillus atrophaeus spores' production by solid-state fermentation (SSF) using sugarcane bagasse as support and soybean molasses as substrate at lab-scale in column bioreactors (forced aeration), plastic bags and Erlenmeyer flasks (aeration by diffusion). Different moisture contents (84%, 86% and 88%; 89%, 91% and 93%) and aeration rates (30mL/min, 45mL/min, 60mL/min and 90mL/min) were studied. The best condition for spore production (3.3x10(10) CFU.g-¹dry matter) in column bioreactor was 80% of initial humidity and no aeration. In Erlenmeyer flasks and plastic bags the best sporulation production reached 1.7 up to 4.7x10(10) CFU.g-1dry matter with 88-93% of initial moisture. The aeration rate had no significant effect on...

The morphology and wall ultrastructure of megaspores and microspores of Isoetes sehnemii that grows in Brazil were analyzed as part of the study of the Isoetaceae present in Southern South America. The observations were performed with light, scanning and transmission electron microscopes. The megaspores are trilete, 350-450μm in equatorial diameter. The surface is reticulate. In section, the sporoderm is 100μm thick including the ornamentation. The wall is composed of a siliceous perispore, which consists of short fused flatten, elements forming a three-dimensional mesh. The spores of Schizaea fistulosa from Chile were studied using light microscopy (LM), scanning (SEM) and transmission electron microscopy (TEM). The spores are monolete and elliptic in polar view. The major equatorial diameter is 71-85 µm and the polar diameter is 54-61 µm. The laesurae are 50-60 µm long and, in some cases, bifurcated. The spore morphology and wall ultrastructure of Microlepia speluncae and Pteridium arachnoideum from the Paranaense Province were analyzed with LM, SEM and TEM and a comparative analysis was carried out. In both species the spores are covered by a three-dimensional network of threads branched and fused, tangentially arranged to the surface, and some free-end threads are also seen. The spore morphology and wall ultrastructure of Sphaeropteris gardneri (Hook.) R.M. Tryon from Brazil were analyzed with LM, SEM and TEM. The spores are trilete with an ornamentation formed of short low ridges with spines in their margins. The exospore is 2.5μm thick, two- layered in section and single or branched channels are present. Se evaluó el efecto de seis concentraciones de conidios de Penicillium digitatum con el objetivo de determinar la concentración óptima que permita 100% de infección en frutos de limón persa en postcosecha. El hongo se aisló de frutos de limón mexicano del estado de Colima, Méx. Cada concentración (1 x 105, 3 x 105, 5 x 105, 7 x 105, 9 x 105, 1 x 106 esporas mL-1), se colocó en heridas de 2 mm profundidad x 1mm de ancho, aproximadamente, hechas en frutos en etapa de madurez de corte. Light is a limiting factor for fern stablishment because it controls germination of light sensitive spores. The aim of this work was to study the effect of light levels on spore germination in two ornamental ferns native to the Atlantic forest, under natural conditions. Cyathea delgadii is a tree fern and Blechnum brasiliense, a subarborescent fern. The effect of light levels was analyzed in April and July/2003, in Florianópolis, Santa Catarina state, Brazil. Sterilized spores were sown in Erlenmeyer flasks containing mineral culture medium with macronutrients, iron, and benomyl 0.01%.
